Background
CD68, also called Scavenger Receptor D1 (SR-D1) and LAMP-4, is
a heavily glycosylated type I transmembrane glycoprotein that belongs to the
LAMP family comprised of five lysosome-specific protein members
including LAMP-1, LAMP-2, dendritic cell (DC)-LAMP (also known as CD208 and
LAMP-3), and brain and dendritic cell-associated (BAD)-LAMP (also known as
LAMP-5) (1, 2). CD68 expression represents the classical immunohistology marker
molecule for human monocytes and tissue resident macrophages (3). It
is mainly associated with endosomal/lysosomal compartment and serves as a
scavenger receptor for oxidized LDL (4). In contrast to other
members of the LAMP family of glycoproteins which are ubiquitously expressed in
all cell types, CD68 appears to have a more restricted, cell-type specific
pattern of expression (5). Human CD68 is expressed as a 354 amino acid (aa) protein, which
includes a 21 aa signal peptide, a 298 aa extracellular domain (ECD), a 25 aa
transmembrane segment, and a 10 aa cytoplasmic domain. Within the ECD, human
CD68 shares 65% and 67% sequence identity with mouse and rat CD68,
respectively. CD68 is a biomarker of rheumatoid arthritis and
Hodgkin's lymphoma (6,7). In addition, CD68 on macrophages binds tightly to
both S100A8 and S100A9 to enhance the cell immunity and influences the
functions of cells through NF-kB/focal adhesion kinase pathway (8, 9).
