Background

Methylation of DNA at cytosine residues plays an important role in regulation of gene expression, genomic imprinting and is essential for mammalian development. Hypermethylation of CpG islands in tumor suppressor genes or hypomethylation of bulk genomic DNA may be linked with development of cancer. To date, 3 families of mammalian DNA methyltransferase genes have been identified which include Dnmt1, Dnmt2 and Dnmt3. Dnmt1 is constitutively expressed in proliferating cells and inactivation of this gene causes global demethylation of genomic DNA and embryonic lethality. Dnmt2 is expressed at low levels in adult tissues and its inactivation does not affect DNA methylation or maintenance of methylation. The Dnmt3 family members, Dnmt3a and Dnmt3b, are strongly expressed in ES cells but their expression is down regulated in differentiating ES cells and is low in adult somatic tissue. Recently, it has been shown that naturally occurring mutations of Dnmt3b gene occurs in patients with a rare autosomal recessive disorder, termed ICF (immunodeficiency, centromeric instability, and facial anomalies) syndrome.

  1. We bought this antibody from your company a few days ago, but I found in your manual, there are two Western blot result of DNMT3b, but their sizes are different? Also, even it shows only a clear single band in your manual, but from our result, the background is very high and lots of non-specific bands, so could you please let me know what's the condition you used for your western blot?
    • Please view our western blot troubleshooting tips. If you are getting many non-specific bands, you may want to check your secondary antibody alone (no primary) to determine what bands may be coming from the secondary antibody. Additionally, I suggest that you use the DNMT3b recombinant positive control Cat NBP2-26582 to help in optimizing your model system as positive control. Also, both mouse and human DNMT3b can have different isoforms, and molecular weights can vary depending on species and isoforms detected/present.
  2. Can you tell me if you sell this ab in PBS only: Anti-Dnmt3b antibody [52A1018] - ChIP Grade NB100-56514. I want it without BSA or azide. 
    • We do have this clone available in an Azide and BSA free format
