![<b>Orthogonal Strategies Validation. </b>Dual RNAscope ISH-IHC: SARS Nucleocapsid Protein Antibody [NB100-56576] - Formalin-fixed paraffin-embedded tissue sections of SARS-CoV-2 infected human lung tissue were probed for SARS-CoV-2 viral RNA (ACD anti-sense specific probe v-nCoV2019-S (848561); Fast Red chromogen, ACD [322360]). Adjacent tissue section was processed for immunohistochemistry using rabbit polyclonal anti-SARS Nucleocapsid Antibody [NB100-56576] at 15ug/mL with 1 hr incubation at 25 degrees Celsius followed by incubation with anti-rabbit IgG VisUCyte HRP Polymer Antibody [VC003] and DAB chromogen (yellow-brown). (A) Poly(I:C) increased autophagy markers in cultured H9c2 rat ventricular cells. (B) Poly(I:C) stimulated autophagosome formation but did not affect autophagic flux. ![<b>Orthogonal Strategies Validation. </b>Dual RNAscope ISH-IHC: SARS Nucleocapsid Protein Antibody [NB100-56683] - Formalin-fixed paraffin-embedded tissue sections of SARS-CoV-2 infected human lung tissue were probed for SARS-CoV-2 viral RNA (ACD anti-sense specific probe v-nCoV2019-S [848561]); Fast Red chromogen, ACD [322360]). Adjacent tissue section was processed for immunohistochemistry using rabbit polyclonal anti-SARS Nucleocapsid Antibody [NB100-56683] at 15ug/mL with 1 hr incubation at 25 degrees Celsius followed by incubation with anti-rabbit IgG VisUCyte HRP Polymer Antibody [VC003] and DAB chromogen (yellow-brown). ![Immunohistochemistry-Paraffin: TLR7 Antibody [NB100-56588] - Analysis of FFPE human brain using TLR7 antibody at 1:20 on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of heat induced antigen retrieval (HIER) using 10 mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching with peroxide block. The sections were incubated with primary antibody for 30 minutes and Bond Polymer Refine Detection (Leica Biosystems) with DAB was used for signal development followed by counterstaining with hematoxylin. Whole slide scanning and capturing of representative images was performed using Aperio AT2 (Leica Biosystems). Neuronal staining of TLR7 was observed.
