Immunohistochemistry-Paraffin: SIRT4 Antibody [NB100-1406] - Staining of paraffin embedded Human Skeletal Muscle. Antibody at 2.5 ug/mL. Steamed antigen retrieval with citrate buffer pH 6, AP-staining.
Western Blot: SIRT4 Antibody [NB100-1406] - Staining of Human Kidney lysate (35 ug protein in RIPA buffer). Antibody at 1 ug/mL. Primary incubation was 1 hour. Detected by chemiluminescence.
WB: Approx. 37 kDa band observed in human kidney and U937 lysates (calculated MW of 35.2 kDa band according to NP_036372.1). In transfected HEK293 transiently expressing SIRT4 a band of approx. 38 kDa is observed. This band is not observed in the non-transfected HEK293. IHC-P: Human pancreas shows staining of the islets of langerhans. IHC: In paraffin embedded Skeletal Muscle shows spotty staining of striated fibres in longitudinal section.

Background
This gene encodes a member of the sirtuin family of proteins, homologs to the yeast Sir2 protein. Members of the sirtuin family are characterized by a sirtuin core domain and grouped into four classes. The functions of human sirtuins have not yet been determined; however, yeast sirtuin proteins are known to regulate epigenetic gene silencing and suppress recombination of rDNA. Studies suggest that the human sirtuins may function as intracellular regulatory proteins with mono-ADP-ribosyltransferase activity. The protein encoded by this gene is included in class IV of the sirtuin family.
