Background

JMJD2A is a member of the Jumonji domain 2 (JMJD2) family and encodes a protein with a JmjN domain, a JmjC domain, a JD2H domain, two TUDOR domains, and two PHD-type zinc fingers. This nuclear protein functions as a trimethylation-specific demethylase, converting specific trimethylated histone residues to the dimethylated form, and as a transcriptional repressor.

Total protein from HeLa, A431, Neuro2A, and PC12 was separated on a 12% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 1.0 ug/mL in 5% block buffer and detected with an anti-rabbit HRP secondary antibody using chemiluminescence. Lysine (K)-specific Demethylase 4A/KDM4A/JMJD2A was also immunoprecipitated by rabbit anti-Lysine (K)-specific Demethylase 4A/KDM4A/JMJD2A antibody NB110-40584. For blotting immunoprecipitated Lysine (K)-specific Demethylase 4A/KDM4A/JMJD2A, NB110-40585 was used at 1 ug/ml. Detection: Chemiluminescence with an exposure time of 30 seconds.
