(B) Flag-tagged PPAR gamma or PPAR gamma (128–505) was co-expressed with HA-MST2, HA-MST2-KD (inactive mutant), Myc-SAV1, and/or Myc-SAV1(1–201) in 293 cells as indicated. Cell lysates were prepared at various times after treatment with 40 µg/mL cycloheximide & then immunoblotted with an anti-Flag antibody. The PPAR gamma protein expression was quantified & the half-life of PPAR gamma protein was calculated. Expression of GAPDH was detected as a loading control.
