Background

Human LILRA3, also known as Leukocyte Ig-like receptor 4 (LIR-4), CD85 antigen-like family member E, Immunogobulin-like transcript 6 (ILT-6) and monocyte inhibitory receptor HM43/HM31,  is a 70 kD Ig superfamily member that belongs to the leukocyte receptor complex/cluster. Mature LILRA3 is 416 amino acids (aa) in length and contains four C2-type Ig-like domains (1, 2). Unlike other LILR family members, LILRA3 is actively secreted (3). Alternative splicing generates additional isoforms with  one containing an extra 18 aa in the signal peptide and/or a 64 amino acid deletion in the C-terminal half of the second Ig-like domain (4). Human LILRA3 shares ~86% aa identity with Chimpanzee LILRA3. Human LILRA3 acts as a soluble receptor for class I MHC antigens. It binds both classical and non‑classical HLA class I molecules but with reduced affinities compared to LILRB1 or LILRB2. It also binds with high affinity to the surface of monocytes, leading to abolish LPS-induced TNF-alpha production by monocytes (5). R&D Systems in-house testing indicates that LILRA3 binds to Angiopoietin-like 7, consistent with the demonstrated functional interactions between other members of these protein families (6).
