Background
Profilin-1 (PFN1) is a widely expressed actin-binding protein that implicates in diverse cellular activities ranging from actin polymerization regulation to control of gene transcription. PFN1 facilitate actin polymerization and cell movement by catalyzing the exchange of actin-bound ADP for ATP, and by liberating actin monomer (G-actin) from its sequestering protein, thymosin-beta4; both processes leading to unidirectional addition of G-actin to elongating actin filaments. PFN1 binds actin for impacting cytoskeletal structures, and interestingly, at high concentrations, PFN1 prevents polymerization of actin, and vice versa. PFN1-PIP2 binding inhibits the formation of IP3 and DG, and PFN1 also inhibits AR/HTT aggregation via G-actin binding. Indeed, PFN1 phosphorylation at Ser-138 site has been shown to reduce its affinity for G-actin that follows inhibition of PFN1/HTT interactions and AR/HTT aggregation. PFN1 has long been considered an essential control element for actin polymerization and cell migration, however, recent data suggests an inhibitory action of PFN1 on motility of certain types of carcinoma cells. PFN1 gene disruption causes gross impairment of development and embryonic lethality in insects/mice models abd defects in PFN1 have been linked to amyotrophic lateral sclerosis 18 (ALS18)
