Background

Autophagy is a process of bulk protein degradation in which cytoplasmic components, including organelles, are enclosed in double-membrane structures called autophagosomes and delivered to lysosomes or vacuoles for degradation. ATG12 is the human homolog of a yeast protein involved in autophagy (Mizushima et al., 1998 [PubMed 9852036]).[supplied by OMIM]

  1. I used ATG12 antibody [NBP2-15501] to detect ATG12 and ATG12-ATG5 complex in rat myocardium tissue lysates by Western blot. In results, I found two bands, one was just below the 55kD, another was right at 15kD, very different from the images on our website. I do not see any article about its molecular weight, can you plaease confirm if the bands are of ATG12 or ATG12-ATG5? May I ask if you got any data on rat myocardium or any references? 
    • Rat's ATG12 has a predicted MW of 15 kDa, so I think you have the right band for that. ATG12-ATG5 complex in rat samples will not run exactly as it does in human (band slightly above 55 kDa). At the moment, we are not aware of any publications that show the ATG12-ATG5 complex on a WB .You may consider using both an ATG5 and ATG12 antibody on their samples to see if the complex is detected with two different antibodies. Alternatively you can use KO lysates or siRNA to knockdown ATG5 or ATG12.
