Background
Thimet oligopeptidase (THOP1) is a thiol-rich enzyme with a HEXXH catalytic motif containing zinc distributed ubiquitously in mammalian cells and it belongs to family of M3 metallopeptidases. THOP1 is an oligopeptidase, as it only degrades short peptides (5-17 amino acids) and because of thiol compounds mediated activation, it is referred to as thimet. THOP1 implicates in degradation of peptides released by the 26 S proteasome and is prone to oxidative oligomerization through formation of interprotein disulfides involving specific Cys residues. Maximal THOP1 activity is maintained by partial S-glutathionylation, which is a mechanism that regulates protein oligomerization and enzymatic activity. Besides exising as a secreted neuropeptide-metabolizing enzyme, it is predominately expressed in cytoplasm and nucleus of neuronal cells. THOP1 is involved in cytoplasmic peptide degradation and cleaves bioactive peptides inside and outside CNS (including degradation of beta-amyloid precursor protein and generation of amyloidogenic fragments). It has also been shown to hydrolyses bradykinin (BK), neurotensin (NT), GnRH (gonadotrophin-release hormone), beta-neoendorphin, alpha-neoendorphin, dynorphin 1-8 and nociceptin/orphanin in vitro. THOP1 activity may be controlled by phosphorylation and/or by intermolecular disulfide bonds leading to inactive oligomers, and it expression is differentially regulated in neuroendocrine and spermatid cell lineages by a transcription factor binding to SRY (sex-determining region Y), CAAT box and CREB (cAMP-response element-binding protein) promoter consensus sequences.
