Background

Nuclear factor of activated T cells, cystoplamic 2 (NFATC2), also known as T cell transcription factor NFAT1, is a member of the calcium-regulated family of transcription factors that regulate gene expression in lymphocytes and control cardiac valve formation (1, 2). The NFAT family is comprised of four members, NFATC1-C4. Human NFATC2 is a 925 aa transcription factor containing several key regions including a calcineurin-binding site, a trans-activation domain, and nuclear localization and export signals. The first three members of this family are thought to be immune-specific forms of NFAT, while NFATC4 has functions in the regulation of cardiac hypertrophy and hippocampal neuronal signaling (3, 4). In resting T cells, the NFAT proteins are present in the cytoplasm in their phosphorylated form. Upon elevation of intracellular calcium concentration and activation of the phosphatase calcineurin, the NFATC proteins are dephosphorylated and translocated into the nucleus where they regulate the transcription of genes for a number of cytokines and surface molecules (5, 6).
