Background

MYOC encodes the protein myocilin, which is believed to have a role in cytoskeletal function. MYOC is expressed in many occular tissues, including the trabecular meshwork, and was revealed to be the trabecular meshwork glucocorticoid-inducible response protein (TIGR). The trabecular meshwork is a specialized eye tissue essential in regulating intraocular pressure, and mutations in MYOC have been identified as the cause of hereditary juvenile-onset open-angle glaucoma.

Myocilin was detected in immersion fixed paraffin-embedded sections of Human Skeletal Muscle using Mouse Anti-Human Myocilin Monoclonal Antibody (Catalog # MAB3446) at 5&#160;&micro;g/mL for 1 hour at room temperature followed by incubation with the Anti-Goat IgG VisUCyte&trade; HRP Polymer Antibody (Catalog&#160;# VC004). Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using VisUCyte Antigen Retrieval Reagent-Basic (Catalog # VCTS021). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to sarcoplasm and sarcolemma in skeletal muscle cells. Flow Cytometry: GRP78/HSPA5 Antibody [NBP1-06274] - An intracellular stain was performed on Jurkat cells with Alexa Fluor 647 conjugate of GPR78/HSPA5 antibody NBP1-06274AF647 (blue) and a matched isotype control NBP2-24893AF647 (orange). Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. Cells were incubated in an antibody dilution of 2 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to Alexa Fluor 647.
