Metabotropic Glutamate Receptor 6 (Mglur6 or Grm6), member of class III of Mglur family/GPCR3 family, is a multi-pass membrane protein localized in cell membrane with restricted expression in retinal inner nuclear layer on dendrites of ON-bipolar cells wherein it interact with effector G protein G0 alpha and act as a receptor for glutamate. An increment of light intensity in retina hyperpolarizes the photoreceptor to set off two opposing signals in OFF and ON bipolar cells, whereas in darkness, glutamate released by photoreceptors depolarizes OFF bipolar cells with hyperpolarization of ON bipolar cells. This ON bipolar cell "sign inverting" cascade involves glutamate mediated activation of Mglur6 leading to G0 alpha stimulation which results in closure of a presumably constitutively active nonselective cation channel TRPM1 (transient receptor potential melastatin 1). mGluR6 activity is mediated by a G-protein that inhibits adenylate cyclase activity and mGluR6 is the only known mGluR that mediates direct synaptic transmission in the nervous system with its role in mediating ON-response in vertebrate retinal ON-pathway. Genetic inactivation of mGluR6 in murine model blocks the ON-response and in humans, mutations in the mGluR6 gene have been linked to congenital stationary night blindness, characterized in the electroretinogram by the absence of an ON response.
