Dynamin-related Protein 1 (DRP1) is known to function in mitochondrial and peroxisomal division and mediate membrane fission through oligomerization into ring-like structure and sever the mitochondrial membrane, through a GTP hydrolysis-dependent mechanism. Down regulation of DRP1 using siRNA and subsequent detection using anti-DRP1 antibodies in various cell lines led to inhibition of the fission machinery in Bax/Bak-mediated apoptosis, by preventing the fragmentation of the mitochondrial network and partially inhibiting the release of cytochrome c from the mitochondria. These observations suggest that the fission of mitochondria is a dispensable event in Bax/Bak-dependent apoptosis influenced by DRP1 (2). The specific Bax/Bak-dependent biochemical transitions in DRP1 properties link the induction of cell death to the changes in mitochondrial membrane dynamics demonstrate that the apoptotic machinery regulates the biochemical properties of DRP1 during cell death (3).
