Application Notes
This NFkB p65 antibody is useful for Western blot where a band is seen ~60 kDa, Immunocytochemistry/Immunofluorescence, and Immunohistochemistry on paraffin embedded sections. Prior to immunostaining paraffin tissues, antigen retrieval with sodium citrate buffer (pH 6.0) is recommended.

Background

NFkB is a pleiotropic transcription factor which exists as homodimeric/heterodimeric complex generated by Rel-like domain-containing proteins RELA/p65, RELB, NFKB1/p105, NFKB1/p50, REL and NFKB2/p52 , and years of research has established that p65-p50 complex is most abundant in various cell types. The dimers bind at kB sites in their target genes DNA and the individual dimers exhibits differential affinity/specificity for acting as transcriptional activators or repressors e.g. p65-p50 and p65-c-Rel dimers acts transcriptional activators. Inside cytoplasm, NFkB complexes are held in an inactive state complexed with NFkB inhibitor or IkB family members which gets phosphorylated through IKKs/IkB kinases in response to different activators, subsequently degraded thus liberating the active NFkB complex which translocates to the nucleus for modulating the target genes. NFkB it has been found as the endpoint of a series of signaling events which are initiated by a diverse array of stimuli associated with critical cellular processes such as inflammation, immunity, differentiation, cell growth, apoptosis and tumorigenesis.
