Application Notes
This PHD1/HIF Prolyl Hydroxylase 1 antibody is useful for Immunocytochemistry/Immunofluorescence, Immunoprecipitation, Immunohistochemistry and Western Blot. In WB a band is seen at 45-50 kDa. In ICC/IF, cytoplasmic and nuclear staining was observed in HeLa cells. IP (PMID: 21282188). IHC (PMID 21877141). Use in Immunohistochemistry-Paraffin and Electron Microscopy reported in scientific literature (PMID 17003483)

In Simple Western only 10 - 15 uL of the recommended dilution is used per data point. Separated by Size-Wes, Sally Sue/Peggy Sue.

Background

HIF prolyl 4-hydroxylases (PHD) are proyl hydroxylase domain-containing enzymes (PHD1/ Egln2, PHD2/ Egln1, PHD3/ Egln3, and P4H-TM) that facilitate physiological reactions to hypoxic stress by modulating HIF1alpha expression levels. HIF-alpha is regulated by hydroxylation, both by a family of PHDs leading to ubiquitination and proteasomal degradation, and by transcriptional inactivation following asparaginyl hydroxylation by FIH (factor inhibiting HIF). HIF Prolyl Hydroxylase 1 (HIF-PH1/EGLN2/PHD1) is localized in the nucleus and interacts with SIAH2, LIMD1, WTIP and AJUBA. In normal oxygen conditions, HIF-PH1 catalyzes the post-translational formation of 4-hydroxyproline in HIF- alpha and hydroxylates a specific prolines found in NODD/CODD of HIF1A, and also hydroxylates HIF2A with preference for CODD site for HIF1A/HIF2A. VHL (von Hippel-Lindau ubiquitination) complex then targets hydroxylated HIFs to facilitate their proteasomal degradation. Under hypoxic stress conditions, the hydroxylation reaction is attenuated allowing HIFs to escape degradation resulting in their translocation to the nucleus, heterodimerization with HIF1B, and increased expression of hypoxy-inducible genes. HIF-PH1 is involved in regulating hypoxia tolerance and apoptosis in cardiac and skeletal muscle, tumor angiogenesis and susceptibility to normoxic oxidative neuronal death.
