This GSK3 beta antibody is useful for Immunocytochemistry/Immunofluorescence and Western blot, where a band is seen approx. 47 kDa. In ICC/IF strong cytoplasmic staining was observed.

Background
Glycogen synthase kinase 3 (GSK3) is a proline-directed serine/threonine protein kinase that was originally identified for its role in glycogen metabolism; however, subsequently it was shown to serve pleiotropic functions, including embryonic development, gene transcription, and neuronal cell function. Mammalian GSK has two isoforms encoded by distinct genes: GSK3A produces a 51-kD GSK3-alpha protein and GSK3B a 47-kD GSK3-beta protein. Phosphorylation of Tyr279/Tyr216 on GSK3alpha/beta is important for enzymatic activity, whereas inactivation of GSK3 can be achieved through phosphorylation of Ser21/Ser9 residues within the N-terminal domain on GSK3-alpha/beta, respectively. Upstream kinases PKA, AKT/PKB, PKC, p90RSK/MAPKAP and p70S6K are known to phosphorylate Ser9 of GSK3-beta. Furthermore, GSK3 is also regulated via interactions with WNT ligand and its receptor Frizzled/co-receptor LRP5/6, and this interaction releases GSK3 from a multi-protein complex formed by beta-catenin, axin, and adenomatous polyposis coli (APC), which prevents GSK3-mediated beta-catenin degradation and induces beta-catenin-dependent gene transcription. GSK3 is one of the few signaling mediators that play central roles in a diverse range of signaling pathways, including those activated by WNT, hedgehog, growth factors, cytokines, and G protein-coupled ligands. GSK3 dysregulation, particularly hyperactivation, with various pathological conditions, including diabetes mellitus, obesity, inflammation, neurological disorders, and tumorigenesis.
