This XRCC3 antibody is useful for Western blot, where a band can be seen at 38 kDa. No cross-reactivity was observed with XRCC2, RAD51, RAD51B, RAD51C, or RAD51D. The antibody is not applicable for Immunocytochemistry/Immunofluorescence.

Background

XRCC3 (X-ray repair cross-complementing group 3) belongs to RecA family and participates in HRR/homologous recombination repair pathway which is an "error free" DNA repair mechanism that utilizes information encoded by homologous sequence to repair double-strand breaks (DSBs), chromosomal fragmentation, translocations and deletions. XRCC3 is required for Rad51 assembly/stabilization and in HRR pathway, XRCC3 protein interacts with Rad51 protein for contributing to chromosome integrity maintainance, thereby preventing mutations which can lead to carcinogenesis. In addition to repairing DSBs, XRCC3 also plays a role repairing wide range of DNA damage arising from carcinogens. Moreover, XRCC3 can control mtDNA copy number under conditions of oxidative stress in the presence of RAD51 as well as RAD51C. It forms a part of a complex consisting of RAD51B, RAD51C, RAD51D, XRCC2 and XRCC3, and has been suggested to forms complex with FANCD2, BRCA2 and phosphorylated FANCG. XRCC3 also interacts with SWSAP1 and ZSWIM7 for executing homologous recombination repair and defective XRCC3 have been linked to susceptibility to breast cancer as well as cutaneous malignant melanoma type 6 (CMM6).
