RSPO-1 (roof plate-specific spondin; also R-Spondin 1 and cristin3) is a 39 kDa, heparin-binding member of the Cristin/R-Spondin family of proteins. These molecules apparently are either retained intracellularly or secreted, where they activate the frizzled-LRP receptor pathway. Mature human RSPO-1 is 27 kDa and 243 amino acids (aa) in length. It contains a cysteine-rich furin repeat domain followed by a type I TSP domain. This form is apparently retained intracellularly. A presumably glycosylated, 30 kDa, 193 aa C-terminally truncated form is secreted and interacts with heparin. The secreted form of human RSPO-1 is 93% aa identical to secreted mouse RSPO-1.
