Background

The family of natriuretic peptides (see MIM 108780) elicit a number of vascular, renal, and endocrine effects that are important in the maintenance of blood pressure and extracellular fluid volume. These effects are mediated by specific binding of the peptides to cell surface receptors in the vasculature, kidney, adrenal, and brain.[supplied by OMIM]

Western Blot: iNOS Antibody [NB300-605] - Analysis of iNOS was performed by loading 20 ug of RAW264 whole cell lysate untreated (left lane) or stimulated with LPS at 1 ug/mL for 16 hours (right lane) and 10 uL of PageRuler Plus Prestained Protein Ladder onto a 4-20% Tris-Glycine polyacrylamide gel. Proteins were transferred to a nitrocellulose membrane and blocked with 5% Milk in TBST for at least 1 hour. The membrane was probed with an iNOS Rabbit polyclonal antibody at a dilution of 1:1000 overnight at 4C on a rocking platform, washed in TBST, and probed with a Goat anti-Rabbit IgG (H+L) Secondary Antibody, HRP conjugate at a dilution of 1:1000 for 1 hour. Chemiluminescent detection was performed using SuperSignal West Pico.

Representative Western blot analysis and quantification of KLF2 (n = 5) and KLF4 (n = 3) levels in whole-cell lysates from nonirradiated (sham) and irradiated HUVECs 4 h and 24 h after exposure to either five fractions of 2 Gy (5 x 2 Gy) or single exposure to 10 Gy. Desmin was detected in immersion fixed frozen sections of mouse skeletal muscle using Human Desmin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3844) at 10 µg/mL overnight at 4 °C. Tissue was stained using the NorthernLights™ 557-conjugated Anti-Goat IgG Secondary Antibody (red; Catalog # NL001) and counterstained with DAPI (blue). Specific staining was localized to z-lines.
