Western blot shows lysates of MCF-7 human breast cancer cell line, HepG2 human hepatocellular carcinoma cell line, and Jurkat human acute T cell leukemia cell line. PVDF membrane was probed with 0.2 µg/mL Goat Anti-Human ERK1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF1879) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). For additional reference, Recombinant Human Active ERK1 (Catalog # 1879-KS) and Recombinant Human Active ERK2 (Catalog # 1230-KS) (2 ng/lane) were included. A specific band for ERK1 was detected at approximately 43 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background

ERK1 (also known as MAPK3, Mitogen-activated protein kinase 3) is a 44 kDa phosphoprotein. It is Serine/threonine proteinkinase which acts as an essential component of the MAP kinase signal transduction pathway. ERK1 was initially isolated and cloned as a kinase activated in response to insulin and NGF. It is expressed in most, if not all, mammalian tissues. Dual threonine and tyrosine phosphorylation activates human ERK1 at Thr202/Tyr204. Within the range used as an immunogen human and mouse ERK1 share 97% amino acid sequence identity.
