Background

Defensins are a kind of endogenous antimicrobial peptide enriched with cysteine/arginine and are widely distributed in internal organs wherein they are recognized as defenders against invasion by pathogenic organisms. Human alpha-defensins (human alpha-defensins 5, -6, and human neutrophil peptides-1, -2, -3, -4) are a class of small antimicrobial peptides with well characterized antibacterial activity against both Gram +ve as well as Gram-ve bacteria, and certain viruses such as such as HSV, HPV, adenovirus and polyoma virus. Human alpha-defensin 5 (HD5) is a 32 amino acids long protein that shares a common structural feature of alpha-defensin family characterized by three-stranded anti-parallel beta-sheets stabilized by three intra-molecular disulfide bridges. HD5 is primarily expressed in small intestinal Paneth cells or epithelial cells of urino-genital tract, and it is found in cytoplasmic vesicle/secretory vesicle or in secreted form. Peptides HD5(20-94), HD5(23-94) and HD5(29-94) are found within tissues among which HD5(20-94) is the predominant intracellular form. In extracellular milieu, peptides HD5(56-94) and HD5(63-94) are found and among these two peptides, HD5 (63-94) is the most abundant form.
