Background

Caspases are evolutionary conserved family of aspartate-specific cystein proteases which are essential part of apoptotic machinery and typically signal in a two-step cascade: 'initiator caspases' with large prodomains (such as caspase-1, -8 and -9) are first recruited into large protein complexes in which they undergo proximity-induced autoactivation and "executioner caspases" such as caspase-3 and -6, caspase-7 which are activated by initiator caspases via freeing of executioner caspases of their short inhibitory prodomain, allowing them to cleave a large set of cellular substrates. Localized in cytoplasm, caspase 7 is a heterotetramer that is consists of two anti-parallel arranged heterodimers, each one formed by a 20 kD (p20) and a 11 kD (p11) subunit, and interacts with BIRC6/bruce. Caspase 7 cleaves/activates SREBPs and poly(ADP-ribose) polymerase (PARP). Its overexpression promotes apoptosis, and it also implicates in inflammation signaling. Therapeutic intervention with caspase-7 has been shown to prove beneficial in conditions where excessive cell death and/or inflammation contribute to diseases e.g. Alzheimer's disease, Huntington's disease, sepsis, rheumatoid arthritis, cancer and NIDDM.

  1. May we ask the MW difference between the active form and the proform of Caspase 8 while performing WB?
    • In treated cells induced to undergo apoptosis, caspase-8 migrates as a 55/53 kDa (pro-form), 41/42 kDa (a cleaved/active or intermediate form), and 18 kDa (active form). The proform (55/53 kDa) is still seen in treated cells because not all cells undergo apoptosis at once.
  2. May we ask if NB600-1343, NB100-56527 and NB500-208 can recognize both active form and profor
    • Product NB100-56527 recognises both pro and active forms of Caspase 8. NB500-208 recognises only the active form, and NB600-1343 has not been tested for specificity at this time.
  3. Does NB500-208 both recognize human and rat active form of caspase 8 (subunit p18 or subunit p10)? Besides that please let me know if NB500-208 also recognize pro-form caspase 8?
    • Caspase 8 Antibody (FLICE 4-1-20) # NB500-208 antibody does not recognize rat samples. It is human specific and in our QC data it was successfully tested to detect the pro or inactive form of caspase at molecular weight 59kd. However, depending upon the extent of extrinsic apoptotic pathway activation in your cells or tissues, you can expect to see bands of the active form also.
