Siglec-1/CD169 Antibody (ED3) [CoraFluor™ 1] Summary

Description
CoraFluor(TM) 1 is a high performance terbium-based TR-FRET (Time-Resolved Fluorescence Resonance Energy Transfer) or TRF (Time-Resolved Fluorescence) donor for high throughput assay development. CoraFluor(IM) 1 absorbs UV light at approximately 340 nm, and emits at approximately 490 nm, 545 nm, 585 nm and 620 nm. It is compatible with common acceptor dyes that absorb at the emission wavelengths of CoraFluor(TM) 1. CoraFluor(TM) 1 can be used for the development of robust and scalable TR-FRET binding assays such as target engagement, ternary complex, protein-protein interaction and protein quantification assays.

Specificity
NB100-65301 recognizes the rat CD169 cell surface antigen, a 185kD molecule expressed by macrophages, predominately confined to lymphoid organs only. Monocytes and granulocytes are negative. No other cell types are positive. The most conspicious property of ED3 is it stains marginal zone macrophages and marginal metallophils in the spleen very strongly. Furthermore, macrophages in (auto-immune) diseased tissues express the ED3 antigen. In healthy tissue no expression occurs. CD169 is a receptor for glycoconjugates containing sialic acid.

Background

The siglecs are a family of membrane bound lectins (of the immunoglobulin superfamily) that bind sialic acid and mediate cell-cell interactions. Family members include sialoadhesin, CD22 and CD33. Sialoadhesin is a macrophage-specific cell adhesion molecule that is expressed on activated macrophages in chronic inflammation and in tumours.
