ChIP: A customer has reported successful usage (no data). WB: Approx. 60 kDa band observed in human lymph node and human spleen lysates (calculated MW of 57.7 kDa band according to NP_000374.1). An additional band of unknown identity was also consistently observed at 48 kDa. This band was successfully blocked by incubation with the immunizing peptide. |

Alternate Names for Autoimmune Regulator/AIRE Antibody
Background
The function of the protein is not well defined, however it contains zinc finger motifs suggestive of a transcription factor. The protein (isoform 1) is localized to both the nucleus and cytoplasm. Three splice variant mRNAs products have been described [1]. The longer AIRE1 mRNA appears to be more abundant and includes exons 1 through 14. Splice variant AIRE2 includes a portion of the non-coding region of exon 1, an alternatively spliced longer exon 8, plus exons 9 through 14. Variant AIRE3 includes the same exon 1-8-9 sequences as found in AIRE2 but utilizes additional alternative splicing in exon 10 that shifts the reading frame such that a stop codon in exon 12 is utilized. The resulting protein products of these splice variants differ significantly. Defects in this gene cause the autosomal-recessive systemic autoimmune disease termed autoimmune polyendocrinopathy-candidiasis-ectodermal dystrophy (APECED).

I am looking into a positive control for Western blotting. We are trying to detect the AIRE protein and are not sure whether the antibody we have is picking up the protein. Are you able to tell me whether the human thymus whole tissue lysate (NB820-59456) has been used to detect for AIRE expression by Western blotting?
- I am not currently aware of any references to the use of our human thymus whole tissue lysate being used to detect AIRE by Western blotting. On looking through the Western blot images for all seven of our AIRE antibodies, I see that these have been generated using lysates from RT-4 cells (bladder), U-251 MG cells (brain), 293 cells (human embryonic kidney), human spleen lysate, liver lysate and tonsil lysate.
