Immunohistochemical analysis of a formalin fixed and paraffin embedded rat brain tissue section using Tyrosine Hydroxylase antibody at 1:5000 dilution. The primary antibody binding to its antigen was detected using HRP anti-Polyvalent ready-to-use kit (Ultratek, 35368) with DAB (brown) and the sections were further counterstained using hematoxylin. Isotype control section was incubated with Rabbit IgG Isotype Control antibody and was processed under the same assay conditions. Tyrosine hydroxylase staining (brown) was observed in the test samples only and the signal was specifically localized to the neuronal cells.

Western blot shows lysates of human adrenal gland tissue, mouse adrenal gland tissue, and rat adrenal gland tissue. PVDF membrane was probed with 0.25 µg/mL of Mouse Anti-Human/Mouse Tyrosine Hydroxylase Monoclonal Antibody (Catalog # MAB7566) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). Specific bands were detected for Tyrosine Hydroxylase at approximately 50-60 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Cells were double stained using the Northern-Lights™ 637-conjugated Mouse Anti-Neuron-specific beta-III Tubulin Monoclonal Antibody (white; Catalog # NL1195V). Cells were counterstained with DAPI (blue). Specific staining of Tyrosine Hydroxylase was localized to cytoplasm of dopaminergic neurons. Tyrosine Hydroxylase was detected in immersion fixed paraffin-embedded sections of human brain (medulla) using Mouse Anti-Human/Mouse Tyrosine Hydroxylase Monoclonal Antibody (Catalog # MAB7566) at 25 µg/mL overnight at 4 °C. Tissue was stained using the Anti-Mouse HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS002) and counterstained with hematoxylin (blue). Specific staining was localized to neurons. Specific bands were detected for Tyrosine Hydroxylase at approximately 52-61 kDa (as indicated) using 5 µg/mL of Mouse Anti-Human/Mouse Tyrosine Hydroxylase Monoclonal Antibody (Catalog # MAB7566). This experiment was conducted under reducing conditions and using the 12-230 kDa separation system. Non-specific interaction with the 230 kDa Simple Western standard may be seen with this antibody.

PVDF membrane was probed with 1 µg/mL of Sheep Anti-Human Tyrosine Hydroxylase Antigen Affinity-purified Polyclonal Antibody (Catalog # AF7566) followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody (Catalog # HAF016). Specific bands were detected for Tyrosine Hydroxylase at approximately 56-60 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Tyrosine Hydroxylase was detected in immersion fixed paraffin-embedded sections of human brain (medulla) using Sheep Anti-Human/Mouse/Rat Tyrosine Hydroxylase Antigen Affinity-purified Polyclonal Antibody (Catalog # AF7566) at 15 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Sheep IgG VisUCyte™ HRP Polymer Antibody (VC006). Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (CTS013). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to cytoplasm in neurons. Staining was performed using our protocol for IHC Staining with VisUCyte HRP Polymer Detection Reagents.
