The unfolded protein response (UPR) is the cellular response to the accumulation of mis-folded proteins in the endoplasmic reticulum (ER). UPR has recently been shown to be important in a number of pathologies involving aberrantly folded proteins. It appears that UPR may be involved in the pathologies of diseases as varied as Diabetes, Alzheimer's and atherosclerosis, in addition to being able to induce autophagy and apoptosis. In response to the accumulation of mis-folded proteins in the ER, a unique signal transduction pathway is activated involving GRP78, CHOP, PERK, ATF6 and XBP-1 proteins, among others.
