MYD88: Fanning Inflammation and Immune Responses

Myeloid differentiation primary response gene 88 (MYD88) encodes a cytosolic adapter protein that plays an essential role in innate and adaptive immune responses. MYD88 protein functions as an essential signal transducer in the interleukin-1 and in Toll-like receptor signaling pathways by acting as an adapter, which regulates the activation of several proinflammatory genes. MYD88 expression has been shown to be a critical factor against parasitic infections conferring resistance to the organisms in which it is expressed. Mice lacking MYD88 expression as detected by flow cytometry and Western blotting using antibodies against MYD88 were found to be more prone to parasitic infections (1).

Knockout mice having loss of epithelial MyD88 signaling resulted in increased numbers of mucus-associated bacteria; translocation of bacteria, suggesting that MYD88 signaling in intestinal epithelial cells is crucial for maintenance of gut homeostasis (2). Clinical significance of MYD88 expression in human ovarian carcinoma was examined by immunohistochemistry using anti-MYD88 antibodies suggesting that MYD88 expression is a significantly poor prognostic factor for ovarian cancer (3). A better understanding of the role of MYD88 expression in disease progression and outcome may be helpful for development of novel chemotherapies for cancer patients.
