Excess body fat is linked to many human diseases, including IRS (insulin resistance syndrome), type 2 diabetes, atherosclerosis and CAD (coronary artery disease). Fat cells were originally thought to be simple lipid storage vessels, but are now known to secrete several metabolically active hormones, called adipocytokines. These include TNF-alpha, IL-6, PAI-1, Resistin, Angiotensin, Adiponectin and leptin.

Human adiponectin accounts for around 0.01% of total plasma proteins. Our antibody catalog includes reagents targeting both the protein and its receptors: AdipoR1, which is expressed predominantly in skeletal muscle, and AdipoR2, which is expressed in the liver. These receptors mediate adiponectin release, which varies between tissue areas.

In both human and experimental animal studies, reduced adiponectin expression is linked to obesity, type 2 diabetes and IRS, with animal studies suggesting adiponectin deficiency as an important factor in the pathogenesis of insulin resistance. In human studies, there is an inverse relationship between adiponectin levels, obesity, type 2 diabetes and metabolic syndrome. In contrast, leptin expression increases exponentially with increased body fat mass. Interestingly, adiponectin and leptin antibody studies in lipoatrophic mice showed insulin resistance could be fully reversed when the proteins were administered in combination, but only partially reversed when each was administered alone.

In 2004, Park et al. probed adiponectin and leptin levels in obese and lean, non-diabetic human subjects, using adiponectin and leptin antibody reagents. It was seen that while leptin was a good marker for adipose tissue, it was not significantly associated with IRS. Reduced adiponectin levels, however, did point to IRS involvement, independent of BMI (body mass index).
