Western Blot
1. Proteins are separated on an SDS-PAGE gel.

2. Following the protein transfer, the membrane is blocked with 5% NFDM in TBS-T.

3. The membrane is then washed.

4. NB 100-181 [anti-hDMC] is diluted 1:1,000 in dilution buffer [5% NFDM in TBS-T] and incubated for 3 hours at room temperature, gently shaking.

5. The membrane is then washed.

6. Secondary antibody [HRP conjugated goat anti-mouse] is incubated for 45 minutes at room temperature, gently shaking.

7. Develop membrane.

NOTE: Mouse testis tissue extracts were used as a positive control for this antibody. MEF 161 and HeLa whole cell extracts were not shown to work in Western analysis.
