Lightning-Link® Rapid Type A Biotin Antibody Labeling Kit Summary

Description
Lightning-Link Rapid Antibody Labeling kits enable direct labeling of antibodies, proteins, peptides or other biomolecules (See protocol for further information) and allow conjugations to be set up in seconds, and used within minutes (less than 20 minutes).

Our Lightning-Link Rapid technology can be used to label small quantities of antibody with 100% recovery. The researcher simply pipettes the antibody or other biomolecule into a vial of lyophilized mixture containing the label of interest and incubates the reaction for only 15 minutes (for more details please watch the video below). As there are no desalting or dialysis steps that commonly interrupt traditional protein labeling procedures.

Features Benefits
Quick and easy to use Save time, no special knowledge required
No separation steps 100% recovery - no antibody/protein loss
Can be used in a wide range of applications Flexible
Freeze dried Ships at ambient temperature, long shelf-life
Fully scalable (10 ug to 1 g or more) Easy transfer from R&D to manufacturing
Stringently QC tested Consistent high quality, excellent batch-to-batch reproducibility
Large number of labels available Experimental flexibility
Reliable: nearly 300 references Successfully used in many fields of research

The attachment of biotin to biomolecules is an important laboratory technique. Biotin binds to the tetrameric avidin proteins, including streptavidin and neutravidin, with exceptionally high affinity, and this interaction is exploited in various applications such as western blotting, immunohisthochemistry and ELISA. Our biotinylation kits are optimised to create high quality conjugates and minimise assay background.

Lightning-Link Biotin Has been optimized for two separate applications:
  • Type A - Intended for assays in which a streptavidin-labeled detection reagent will be used.
  • Type B - Optimised for assays in which the biotinylated protein is captured by streptavidin immobilized on a surface (ie plates, nitrocellulose, magnetic beads etc).

Applications/Dilutions

Application Notes
By circumventing the desalting or dialysis steps that commonly interrupt traditional antibody conjugation procedures, LightningLink technology can be used to label both small (e.g. 10 ug) and large quantities of primary antibodies with ease. Batch-to-batch variation upon scale up is minimal as the process is so simple, and recoveries are always 100%. This kit is supplied with 3 vials, each suitable for labeling up to 200 ug of antibody.

FAQs for Lightning-Link (R) Rapid Type A Biotin Kit (370-0010). (Showing 1 - 3 of 3 FAQs).

  1. Hi, I'm interested in buying your Lightning-Link Biotin labelling kit. I would like to couple biotin to an antibody, which would bind to an antigen in a sandwich assay format. Then streptavidin-coated nanoparticles would be used as the detection scheme. Which type is best for this purpose, type A or type B? Since Type A is for when Strep is used as a detection reagent, but Type B says its best for when Strep is immobilised to the suface of nanoparticles. In my case it is both.
    • Type B would be the preferred kit.
  2. Whats the difference between Lightning-Link (R) Rapid Type A and Type B for Biotin antibody labeling?
    • Type A - Intended for assays in which a streptavidin-labeled detection reagent will be used.Type B - Optimised for assays in which the biotinylated protein is captured by streptavidin immobilized on a surface (ie plates, nitrocellulose, magnetic beads etc).
  3. What about Biotin is different for Type A and Type B 370-0010 and 371-0010?
    • The actual biotin that is used in the label is the same biotin. Biotin A has been optimized particularly for performance; it should be used when the biotinylated antibody will undergo a wash step before coming into contact with streptavidin. For example in a sandwich ELISA, free label is not a problem because a wash step will remove unbound Biotin before you add streptavidin. Biotin B has been specially developed to further minimize excess left over biotin, and should be selected if the biotinylated antibody will NOT undergo a wash step before coming into contact with streptavidin. e.g. if the biotinylated antibody will be captured on streptavidin-beads. Free, left over biotin is a problem here because it is not washed away. It is captured by the beads and so affects the signal.
